Cytochrome c (Cyt.c) in human plasma was characterized by electrofocusing on polyacrylamide gels. Using isoelectric focusing in the first dimension, a multicharge complex of Cyt.c was separated from unknown peptides and polypeptides. The Cyt.c (8.5 +/- 0.2 kDa) was the major protein in human plasma. A highly selective affinity ligand, dibromomaleimide, was developed to prepare native Cyt.c. A refinement of the spectral parameter assignment for the Cyt.c in the literature is presented. The high resolution Edman sequencing of the amino terminal sequence of Cyt.c in human plasma showed a sequence of FSDK, which corresponds to the sequence of mature Cyt.c in bovine plasma.The risk of complications in microvascular reconstruction. A retrospective study of 100 consecutive free flaps was undertaken to assess the risk of developing major complications following the harvesting of a free flap and to evaluate the reliability of different vascular regions as donor sites. A total of four different donor regions were used: the superficial inferior epigastric artery (30 flaps); the superior thyroid artery (27 flaps); the medial sural artery (30 flaps); and the lateral circumflex femoral artery (13 flaps). Although the donor site was generally considered to be the greatest problem in free-flap surgery, overall complications were much less frequent than might have been predicted from previous studies, which had included all major complications. The superficial inferior epigastric artery flap showed the least serious complications, with a minor or major complication rate of 25 per cent.
